
Hydrogen supplementation in medicine, both at a pre-clinical and clinical trials level have shown a lot of promise in treatment or preventive effects in the last two decades or so, for a wide variety of ailments ranging from cancer to rheumatoid arthritis, dermatitis and recovery from ischemia with so far, no negative side effects being reported. The data suggests that at least one of the biological targets of hydrogen must have a very high affinity for it.
H2 has unique advantages in clinical applications. It effectively penetrates bio membranes to reach cell nuclei and mitochondria1. Hydrogen supplementation can be delivered safely to patient by multiple means, 1-4% inhalation, Hydrogen rich water (up to 10ppm) or hydrogen rich saline. The mode of action of H2 on the body identified so far is that H2 act as direct and indirect antioxidant especially by scavenging the infamous Hydroxyl radical2 and increasing the activity of both superoxide dismutase and catalase. Furthermore, H2 supplementation is shown to increase ATP production by the mitochondria by more than 50%3 while suppressing the production of superoxide by the first complex of the respiratory chain4.
Hydrogen and Cancer
Hydrogen supplementation have shown great promise in the field of oncology both in cultured cell model, animal model and clinical trials. In cells culture models of Human tongue cancer, human fibrocarcinoma5,6, human cervical cancer and human soft tissue cancer7 H2 supplementation blocked cancer cell proliferation and decreased ROS production. In experimental animals model H2 demonstrated benefits against UV-induced skin cancer8, Ionizing radiation induced thymic lymphoma9, high-fat diet-induced liver cancer 10, mouse colon cancer11, human lung cancer12, Rat and human glioma13, and Human endometrial cancer14.
Using oxyhydrogen gas inhalation, case study have demonstrated impressive potential in the treatment of gallbladder cancer, lung cancer with multiple metastases, and in allievating hearing loss in nasopharyngeal carcinoma15. Hydrogen treatment also restored exhausted CD8+ T cell in colorectal cancer16, while patient with stage IV and V non small cell lung cancer inhaling oxyhydrogen for two weeks saw they exhausted immune cell all reactivated to normal level17.
Hydrogen and Ischemia
Ohasawa et al.2 first reported the beneficial effect of hydrogen upon cerebral infarction. Since then, many studies showed that hydrogen supplementation have positive effect on ischemia/reperfusion injuries18–52, nervous system injuries 53–68 and organ transplants69–87.
Hydrogen as an Anti-Inflammatory
Molecular Hydrogen has proven to act as a powerful antioxidant in biological systems directly and indirectly.
- Directly, by scavenging (neutralising) the most potent Reactive Oxygen species, such as the infamous hydroxyl radical2
- Indirectly, by boosting the production of enzyme parts of the natural antioxidant system such as catalase and superoxide dismutase88–92.
Molecular Hydrogen also appears to potentiate the normal functioning of the mitochondria by restoring the use of oxidative phosphorylation93, but the action of Molecular Hydrogen concerning inflammation doesn’t stop there.
Molecular Hydrogen also seems to decrease the relative number of M1 compared to the healing-promoting M2 macrophage94,95, probably by reversing the metabolic switch or promoting its reversal. In doing so, Molecular Hydrogen avoids an imbalance between M1 and M2 macrophages, helping to bring about healing by decreasing inflammation.
Finally, Molecular Hydrogen seems to be able to downregulate NF-κB58,96–107 providing with a new therapeutic route as an adjuvant treatment for all inflammatory dis-ease, and within the current pandemic could help mitigate the risk of cytokine storm associated with severe COVID108,109.
